Abstract


The purpose of this work is to observe the effect of chlorpromazine on the metabolism of brain and other organs which is being used in many fields of medicine these days.
Studies were made on the metabolism of the various parts in the central netvo~as &,--stem of the rat, e.g., cerebral cortex, cerebellum, diencephalon, medulla oblongata and spinal cord, also on the metabolism of the some visceral organs, e.g., liver, kidney, lung, heart and spleen. The oxygen uptake were examined in vitro by Warburg´s manometric method. Rats weighing 200 to 300 gm were used for the experim ents. Results are as following:
1. The clinical doses of chlorpromazine, i.d., largactil made in France, reduces the oxygen consumption of all parts above metioned in the central nervous system except for the medulla, while, among those visceral organs, decreases of the Q02 were unnoticeable in those organs except for the lung of which Q02 decreased significantly.
2. The results obtained from large doges of chlorpromazine were compared with those from the clinical doses. Larger concentration of chlorpromazine in the medium brought about more marked inhibition of oxygen uptake of the brain stem than other regions of the central nervous system. The inhibitory effect of chlorpromazine on oxygen uptake in the brain stem may be as great as increasing concentration of it. The decrease of oxygen uptake of both lung and kidney were remarkable, while, in the other organs, shows no significant changes.
