Abstract


There are many investigators who have reported that the effect of inhibiting factors on tissue growth, their tissue specificities, and species-specificities. Herbut reported that such a substance had been extracted from Serum and plasma.
Also, Kim, Kim, Lee & Suk, and Hong reported that inhibiting factors extracted from rat liver by Chin´s method, had tissue specificities.
Although many thetories thus far Lacks Uniformity as to it´s species-specificities.
And yet, no Reports Concerning the effect of inhibiting factor extracted from tissue are available.
Present Report is the Result of the study concerning Tissue Growth inhibiting factors extracted from Liver of various Animals, by Chin´s method, and observation of species-specificities.
In regard to the inhibiting reaction among tumor transplanted animal and the effect on the mitosis of Liver and Kidney of normal various animals by injection of Tissue Growth inhibiting factors.
Materials and Methods.
1) Ehrlich Ascites Cancer and N-F Sarcoma were used. The method of tumor Transplantation followed close by that of Hang´s method.
2) Tissue factors were prepeared from various animal´s Livers such as mouse, Qunia Pig, Rat, Rabbit, Dog, and Cow, etc.
Specimen was centrifuged in Model P.R.I. international centrifuge at 15,000/r.p.m., dehydrated ethylalcohol was added to the supernatant and the Mixture was kept in the refrigerator for 24 to 48 hours.
The resulting precipitant was resuspended in small amount of distilled water and subjected to paper electrophoresis in Sodium barbital and Acid barbital Buffer system at PH 8.6. The slow-moving fraction was treated with Trypsin in 37℃ incubator under PH 8. The liquid was shifted in to a cellophane tube and dialyzed in the refrigerator for 72 hours against distilled water. The Substances in the cellophane Tube were removed and precipitated by adding the same volume of aceton.
The sediment was then ready for injection as a 10% solution.
3) 1st Experiment.
The injections started on the day of the tumor Transplantation of the mice.
The daily doses per animal consisted of 0.1 cc 0.2 cc. 0.3 cc of 10% Solution of Tissue Factors. To Observe the Species-Specificity, tissue factors extracted from Various Animals (homo, & Hetero) were injected to 6 groups of tumor implanted Mice, and the effect was measured by the size of tumor directly 3every days. On the 18th day Sacrificed and the Tumors were removed and weighed. The growth of the Tumor was measured by one half of the value of the longest length and longest width, and was compared with control homogenous, and heterogenous group.
To observed the Species-specificity among the control, homegenous, and heterogenous, 1.0 cc of 10% solution of tissue factor of Liver extracted from Rabbit and Rat was injected intraperitoneally to the Rabbit and 0.1 cc of solution of tissue factor of Liver extracted from Rabbit and Rat was injected intraperitoneally to the Rat every day. When animals were Sacrifiecd after injections of three, six, nine, twelve, fifteen, and eighteen successive days respectively. Tissue slices were cut from the Liver and Kidney in appoximately 2 ㎜. thicknesses, and fixed in carnoy´s fluid for three hours, and stained with hematoxyline and eosin. The Mitotic cells of the Liver as well as the Kidney were counted for 100 Fields on each slide with the help of 43 x objective lens and lox Oculars, and Compared with coutrols.
5) Results.
① The effect of Tissue-Growth inhibiting factor to Tumor transplanted animals showed more severe inhibition on Homogenous Group than on Heterogenous Group.
② The effect of Tissue-Growth inhibiting factor to Mitosis on Liver and Kidney of various animals showed more pronounced inhibitory reaction on Homogenous than Heterogenous.
③ The species-specificity can be recognized by observing the reaction of Tissue Growth inhibiting factors.
