Abstract

The influences of cortisone acetate on the fracture healing and the distribution of S^(35) were studied in the rats. Twenty four mature healthy rats were divided into 2 groups, 12 in each, one the control and the other the experimental animals. After the artificial fracture of the femur under ether anesthesia, the control group received 0.2 ㎖ of physiological saline on everyday for 10 days intramuscularly, and the experimental group, cortisone acetate in dose of 30 ㎍ instead of saline. S^(35) was given to the both groups as dated in the Report 1, and the micro-, and macroautoradiography in accordance with the special
metachromic staining with toluidine-blue and eosin were done. Following were the results:
1. A large amount of S^(35) and mucopolysaccharide appeared in the fracture sites during the healing process.
2. The mast cells mostly appeared in the periosteal layer, hence were considered to play an important role in the fracture healing.
3. In the cortisone treated animals, the growing plate of the epi, and diaphyses became thinner and S35 uptake was also diminished.
4. A large amount of the irregular fibrous tissue was also observed in the fracture sites, and most of the hypertrophic chondrocytes disappeared while the chondroid matrix increased.
5. Cortisone acetate appeared to increase the chondroitin mucopolysaccharide in the fracture sites, epi-, and diaphyses, but to suppress the differentiation of the chondrocytes and eventually the bone growth.
