Effects of Acute Ethanol Intoxication on Some Metabolic Activities of Rat Liver and Cerebral Cortex

Abstract


Single dose of 50% ethanol was orally administered to rats(1.5 ㎖ per 100 gm body weight) and the rates of tissue oxygen consumption(QO₂), carbon dioxide production(QCO₂), respiratory quotient(RQ) and succinic dehydrogenase (SDH) activities of the liver and cerebral cortex were studied by the Warburg´s manometric method. The nucleic acid(RNA & DNA) contents of the liver and cerebral cortex were also studied by the method of Schmidt, Schneider & Thannhauser and the contents of blood lactic acid were measured with the blood obtained by cardiac puncture.
Intervals of experiment were 1, 3, 6, 12 and 18 hours and the results obtained are summarized as follows:
1. The oxygen consumption rate(Q0₂) of the liver increased above the normal at 1(129%) and 3 hours (126%) following the ethanol administration and at 6 hours a decline of QO₂ to the normal value was observed with the consequent decrease till 18 hours(87%). The QO₂ of the cerebral cortex showed an increase at 1 hour (116%) but as the time progressed after the ethanol intubation the QO₂ decreased steadily to 86% at 12 hours with the continued decline up to 18 hours.
2. The carbon dioxide production rate (QCO₂) of the liver increased considerably at 1 hour (153%) but began to decrease from 3 hours after the intubation reaching the normal value at 6 hours with a continued decrease of QCO₂ till 18 hours(84%).
The QCO₂ of the cerebral cortex increased to 124% at I hour following the ethanol administration but a gradual decrease of QCO₂ was noted as the time progressed, the value at 12 hours being 85% with the continued tendency of decrease till 18 hours after the intubation.
3. The respiratory quotient(RQ) of the liver was found elevated at 1 hour but a gradual decrease was noted thereafter till 12 hours to the normal level after the intubation. The RQ of the cerebral cortex showed a tendency of a slight increase at one hour after the ethanol intubation.
4. The succinic dehydrogenase(SDH) activity of the liver was increased to 124% at 3 hours, followed by a gradual decrease till 18 hours(82%) after the intubation.
The SDH activity of the cerebral cortex also showed an initial increase at 1 hour(136%) but a gradual decrease was noted till 18 hours(78%) following the ethanol intubation.
5 The ethanol administration showed no significant effect on the RNA contents of both liver and cerebral cortex while the DNA contents were found to decrease as the time progressed following the intubation up to 18 hours(69% & 68% respectively).
6. The contents of blood lactic acid was elevated considerably at one hour after the ethanol intubation.
