Histochemical Study on the Effect of Calciferol on Mucopolysaccharides in the Rats´ Oral Mucous Membrane

Abstract


Twenty International Units of Calciferol in doses of gram per day were given to rats and the amount of mucopolysaccharides in the oral mucous membrane was determined after 1, 2, 3, 5, 7, and 10 days. The tissue excised from the oral mucous membrane was treated in the Carnoy´s fixer and cut in paraffin block. Hematoxylin-Eosin stain was done for histological observation. Polysaccharides was determined by the periodic acid Schiff reaction, while acid mucopolysaccharides by metachromasia of Ohno´s (pH 2.5, 4.1, 7.0) and Pearse´s stains.
The results were as follows:
1. There existed no remarkable differences in PAS reaction of epithelium and connective tissue of the oral mucous membrane between control and calciferol groups.
2. Metachromasia in cytoplasm and intercellular substance of stratm spinosum and stratum granulosum was slightly increased after 2 and 3 days of calciferol administration and returned to the level of control after 5 days. This metachromatic substance appeared to be hyaluronic acid.
