Abstract


It has been debated whether the protracted therapeutic administration of adrenocorticosteroid hormone does cause delay in healing of fractures. This paper represents the study of long term effects of prednisolone upon - the experimental fracture healing of the ribs in rabbits. Daily dosage of prednisolone, 0.5 mg./kg. body weight, was administered, or equivalent to the therapeutic dose in human patients. For this study, 110 healthy rabbits were used, 25 being control(Group 1 ) and 40 being prednisolone treated rabbits with the experimental fracture in ribs(Group 9 ). The remaining 45 in Group II were treated with prednisolone 4 weeks prior to the induced experimental fracture. Following the fracture, daily administration of prednisolone was maintained to the date. of sacrifice. In all of these groups, 5 rabbits formed a unit, which were sacrificed every 7 days.
The following, roentgenographic, gross and microscopic findings were collaborated in evaluating the fracture healing.

It was found that the prednisolone treated animals showed evidences of delayed fracture healing. It took roughly one and half times longer to attain bone union in Group I and twice as long in Group If as compared with the Control Group. It was, however, noted that the formation of primary callus on the fracture site was not delayed in the prednisolone treated animals.
Furthermore, the remodeling process of bone following fracture union is remarkably delayed among those treated with prednisolone. It was also confirmed that the prednisolone treated animals developed osteoporosis in skeleton, particularly in vertebrae and ribs.
In evaluating these findings, microscopic examination is more reliable than those evaluated by gross and roentgenographic examination.

Supplementary studies of serum calcium and phosphorus were made.
Early depression of serum calcium concentration was noted and the serum phosphorus level was persistantly depressed among those prednisolone treated animals. It was further noted that the serum alkaline phosphatase level did not increase, but rather decreased by prolonged prednisolone administration.
