Abstract


The effect of sex hormones on the metabolism of acetate-l-C79 in the femur of mouse was studied. Sex hormones, estrogen, androgen and progesterone, were administered in dose of 0.1 mg per gram of body weight. Normal saline was used as control. Animals were divided into two major groups for experiment: one in vino and the other in vitro. Each group was subdivided into four groups; control, androgen, estrogen and progesterone. Sex hormone was injected subcutaneously at an interval of 24 hours for three successive days, and simple fracture of the left fle nur was induced on the day of last administration of each hormone. Two days after the fracture the animals were sacrificed. Both femurs, one normal and the other fractured, were recovered separately for study. In in vitro experiment, two days after simple fracture of the left femur both femurs were taken out separately as in vivo experiment, and were immersed for 24 hours in the solution impregnated with each of the sex hormones. In each subdivision each femur was pulverized and was homogenized with an incubation medium after the Frantz´ method. Incubation mixtures were incubated for two hours at 38C. After incubation, the CO production rate, specific activity of respiratory COz, and incorporation rate of acetate-1-04 into cholesterol and saponifiable fraction were measured. In addition, the cholesterol in bone tissue was measured by the Zack method.
From the results of the experiment the following conclusions are obtained:
1. The CO2 production rate and specific activity of respiratory CO, in bone are increased by fracture and by administration of sex hormones, both in vivo and in vitro, group and are remarkably elevated in the androgen group, the order of increase being the androgen group, estrogen group, progesterone group and control group.
2. The content of cholesterol and the incorporation rate of acetate-l-C14 into cholesterol in the bone tissue are also increased by fracture and sex hormone. The maximum value is obtained in the androgen group.
3. The amount of saponifiable fraction and the incorporation rate of acetate-1-C14 into this fr-
action are decreased by fracture and by administration of sex hormones. Both are remarkably
decreased in the androgen group. The order of decrease, however, was the same as in the others. T he summary is that sex hormones increase the amount of cholesterol, oxidation of acetate
and synthesis of cholesterol and decrease the synthesis of saponifiable fraction in bone, espe-
cially tie fractured one.
