Abstract


Healthy adult mongrel dogs (36 dogs) weighing 10 to 18 kg (mean 12. 9 kg) were used to study the effects of chlorpromazine (CPMZ) and/or hypothermia after acute bleeding. Rapid bleeding was carried out via femoral artery into a sterile reservior, and the bleeding volume was equivalent to 3% of body weight of each dog. The blood that had remained in the reserviors was reinfused via femoral vein as fast as possible by gravity.
Thirty-six dogs were divided into four groups. 4
Group I. Control group: Six dogs were anesthetized, bled and then reinfused.
Group II. Chlorpromazine group: A: Five dogs were -administered with 5 mg of CPMZ intramuscularly an hour before the bleeding. B: Five dogs were administered with 50 mg of CPMZ intramuscularly an hour before the bleeding. C: Five dogs were administered with 0. 5mg/kg of CPMZ via femoral vein with the blood reinfused.
Group III. Hypothermia group: (esophageal temperature; 31°±1°C) Five dogs were subjected to hypothermia alone 30 minutes after bleeding.
Group IV. Hypothermia and chlorpromazine group.
A: Five dogs were given with 5 mg of CPMZ as Group I, A.
B: Five dogs were given with 50 mg of CPMZ as Group Il , B.
In each group, cardiac and pulmonary functions were studied and results were summarized as follows:
I. When chlorpromazine was administered to the animals either 60 min. (I. m.) before or 90 min. (I. v. ) after the acute bleeding, the drug may exhibit the action of decreasing of total peripheral resistance even though blood pressure further declined as compared with control group.
2. Hypothermia. alone or hypothermia with CPMZ pretreatment had detrimental effects on hemorrhagic shock in dogs.
3. We believe that CPMZ is a useful drug to ameliolate the tissue perfusion by minimizing the degree of vasoconstriction which is always accompanied with hemorrhagic shock.
