Effects of the Biliary Duct Ligation on the Gastroduodenal Mucous Substances in Rabbits

Abstract


In order to study on the morphological and histochemical changes of mucous substances in the rabbit gastroduodenal mucosa following bile duct ligation, this experiment was performed.
Healthy adult male rabbits weighing about 2kg body weight were divided to normal and bile duct ligated groups. Bile duct ligation was aseptically done under sodium nembutal anesthesia and animas were kept in room temperature. Three animals in each group were sacrificed successively by ether inspiration at 3 days, 1, 2 and 3 weeks after the operation.
Specimens of stomach and duodenum were fixed with buffered 10% neutral formalin solution, embedded in paraffin wax, sectioned 6 micra thickness, and stained with hematoxylin-eosin, alcian blue (pH 2.5) PAS (periodic acid-Schiff reaction), colloidal iron-PAS and, according to necessities comparing with staining reactions, alcian blue (pH 2.5), azur A (pH 1. 5, 3. 0, 4. 5) or alcian blue (pH 2.5)-Kernecht-rot stains.
Obtained results are as follows:
1. In normal gastric mucosa of rabbits, thin purple mucus coating is seen over the superficial epithelium, and the surface gastric epithelium reveals from purple to red in variable degree. Alcianophilia predominates in the superficial convolutions and strong reactivity with PAS is found in the foveolar walls. The mucous neck cells show red tint and possibily secrete neutral mucosubstances which differs from the surface epithelium.
The parietal cells shcw week red in the lumiral cyieplasm in the upper portion of gastric glands. The chief cells in the upper portion of the glands reveal alcianophilia.
2. In the gastric muccsa of bits duct ligated rabbits, the gastric foveolae are slightly enlarged temporarily. Blue tint in the chief cells and red one in the parietal cells are increased deep in the glands of the pyloric portion and the body of the stomach.
At one or two Weeks after the ligation, atrophy of the niuco5-a, especially of foveolar epithelium are observed. The columnar epithelial cells show red, and the parietal cells and chief cells which have stainable mucosubstances in the supranuclear portion of their cytoplasm are found in the middle or lower portion of the glands. In this stage, decrease of alcianophilia and relative increase of PAS positive reaction are observed. At three weeks after the ligation, morphological return to the control group except increase of red density of the parietal cells in the pyloric portion is noticed.
3. In the erosed gastric mucosa or the mucosa near the ulceration, depletion of alcianophilia of the chief cells associated with hyperplasia and ballooning of parietal cell cytoplasm occur and this phenomena indicate morphologically destroy of protective balance of the mucosubstances of the gastric mucosa.
4. Blue or purple mucous coating over the columnar cells of villi, blue granules in the luminal cytoplasm of Brunner´s gland cells, and mucins of the goblet cells which reveal various colours from blue to violet according to level are observed in the duodenal mucosa of the normal rabbit. Quantitatively proximo-distal distribution of the goblet cells in the duodenum is noticed and the goblet cells with blue hue are usually located in tips or sides of villi.
5. Duodenal erosion or ulcers, especially in the mucosa near the ligated bile duct, are found in all of experimental rabbits. Alcianophilia of the Brunner´s gland is decreased in the third day after bile duct ligation, and increased during one or two weeks groups. Number of goblet cells is increased in this stage and their staining variability from blue to violet, which indicate hyperactivity of the cells is also observed. Alcianophilia is returned nearly within the normal limits at the three weeks after the ligation.
6. In or around the duodenal mucosa with erosion or ulceration, depletion of alcianophilia of the Brunner´s gland cells and the goblet cells and interruption of the mucus coating over the surface epithelium are observed. This figures indicate destruction of protective role of mucous substance. in the duodeum.
