Abstract


Neumann´s study on spore germination of anaerobes provided a new method for the observation of spore germination. He made continuous observations by using solid media.
Some writers (Burchard, Gottheil, Neide, Salle and Lamanna), believe that the mode of spore germination is constant and, therefore, is valuable in distinguishing one kind of bacillus from another. Others (Grethe, Henrici, Gray), believe that the mode of germination is variable for the same bacillus.
According to Knaysi, the mode of germination is very constant for some species but is quite variable for other species.
Lamanna classified the modes of germination as follow:
1) Spore germination by shedding of the spore coat.
a. Equatorial germination.
b. Polar germination.
c. Comma-shaped germination.
2) Spore germination by absorption of the spore coat.
By making a one cell culture on film media the following observations and advantages were noted:
1. The mode of spore germination of anaerobes can be observed continuously.
2. Photographs can be made easily by using dark field illumination.
3. The method is easily utilized.
4. The percentage of spore germination of anaerobes is less than that of aerobes. As Neumann reported, however, it is more than was earlier reported.
5. Although there is no definite relationship between the ages of spores and the percentage of them germinating, there is a tendency for a greater percentage of older spores to germinate.
6. Spores of anaerobes germinate by shedding their spore coats at the poles i. e. by polar germination.
7. From one spore only a single young cell germinates.
8. The direction of growth of the newborn vegetative cell is parallel to the axis of the spore.
9. The original spore membrane, keeping its refractive property, is visible for a long time after germination.
