Abstract


Three modes of spore formation are reported in the literature: 1) upgrowth from a tiny refractile granule; 2) fusion of numerous sporogenic granules; and 3) condensation of protoplasm.
Growth from a single granule was first reported by Koch. Although some support fir this theory is found in the literature, it is now generally regarded as erroneous.
Spore formation by fusion of numerous granules was first reported by Ernst and Babes. This theory is still upheld by some bacteriologists; however, many of the older as well as the more recent works are opposed to this theory. We now know that the so-called sporogenic granules of earlier writers are volutin granules, fat bodies, etc. and that they occur in many species which do not form spores.
The majority of reports in the literature dealt with only aerobic spore formation and most of the methods seem to be inadequate for continuous observation of growth.
The following results were obtained on Cl. tetania, botulinum, sporogenes, histolyticum, butyricum and sphenoides by making a one cell culture on 1% lactose agar film media (pH 7.0) and then by making continuous observations of their mode of spore formation.
1) The mode of spore formation of anaerobes could be observed continuously by the method described above.
2) Granules were not observed during the period of spore formation.
3) The width of a primordium of a sporangium is greater than that of a vegetative form.
4) The spore resulted from a condensation of the substance contained in the spore primordium.
