Abstract


For the paper electrophoretic separation of the serum protein fractions we need some special equipments, so it would be very useful if such separation is possible by simple paper chromatography. It has hardly been seen on, literatures about the paper chromatography of raw serum proteins.
First of all, as the result of preliminary test for the search of proper solvents, it was found, in general, that serum protein could not be developed by organic solvents. As the result, the author used the Michaelis´ ammonia and barbital buffers of various pH. Toyo No. 51 and Whatman No. 4 filter paper and the ascending method were employed, and the serum protein on the filter paper was identified by Durrum´s bromphenol blue staining and the author´s eosin staining method.
It was observed, in this experiment, that regardless of the pH and the kinds of buffer, the serum ascends very well (in every cases Rf is over 0.8), but no fractional separation could be seen in every paper chromatograms, and even more the Rf values were lack in constancy and reproducibility.
Consequently, it can be said that in the paper chromatography of the serum by aqueous buffer solution, the serum are not developed by the counter current distribution, but rather simply proceeds by the stream of solvent as it infiltrates up the paper overcoming the van der Waals´ adsorptive forces between the paper and the serum. Accordingly, it might be said that paper chromatographic separation of the serum protein fractein is not feasible.
