Abstract


There are many reports investigating the effects of orotic acid on the fatty liver and lipid metabolism. It was reported that large doses of orotic acid, 1 % or 1.5 % of diet, could induce fatty liver on rats, and also that 0.01 % or 0.1 % of orotic acid in diet enhances the metabolism of some nucleotides and do not affect the lipid metabolism in experimental animals. The author was to investigate the effect of orotic acid on the liver damaged by CC14 injections and long term administration of red pepper, and to evaluate the effects of "Orotsan" , preparation of orotic acid, on the damaged liver induced by CC14. Material
Thirty seven male rabbits fed with normal diet composing of 200 gm of bean curd refuse and 50. gm of wheat flour were used in this experiment. The experimental groups were divided as following;
Group A, 9 rabbits were injected with 0.05 c. c. Of CC14 per kg. of body weight twice a week, for 6 weeks with the administration of 30 mg of orotic acid per day by
Month. Group B, 9 rabbits were injected with CC14 like Group A´s and served as controls to Group A.
Three animals in each group were sacirficed after the continued injections of CC14 for two weeks and served as histologic examination.
Group C, 5"rabbits were fed with 10 gm. of red pepper plus 10 gm. of orotic acid for 8 months.
Group D, 5 rabbits were fed with 10 gin. of red pepper only and served as controls to Group C.
Group E, 9 animals were injected with 0.05 c. c. Of CC14 per kg. of body weight for five months. After surgical biopsy of the liver, the injection was stopped. After the cessation of the CC14 injection in Group E, five animals among nine were fed with ordinary diet plus 2 tablets of Orotsan for 1 month, and the remainder were fed with ordinary diet only for the control to aforementioned group.
The results observed are summarized as follows:
1) The administration of orotic acid to CC14-injected animals inhibits the intensity of CC14 toxicity on the liver.
2) The administration of orotic acid in diets inhibits the toxicity of red pepper to the liver.
3) The administration of Orotsan romotes the recovery from the live r damage induced by CC14.
