Abstract


S-R form variants of Brucella strains were obtained after several successive cultivation of them in antiserum containing broth.
Tn, addition to the routine procedure, - such as agglutination by antiserum and acriflavine dye solution, heavy metal ion reactions were employed for the classification of the variants and each classified mutant was investigated as to its immunogenicity and virulence with the following results.
1. S-form: the normal and standard colony which did not react at all in any agents except for tl~´antiserum.
2. R-form: the extremely- dissociated form which agglutinated spontaneously even in saline.
3. SR-form: the intermediate form between S-and R-forms; in antiserum a similar pattern to S-form was observed, while in acriflavine, lead acetate, and manganese chlorid solutions a moderate Mme,: .reaction occurred.
4. Virulence test revealed that SR-form was as attenuated as R-form.
5. Immunogenicity of SR-form to mice turned out as good as that of ether-killed or phenol-killed vaccine, while that of R-form was negligible.
6. The characteristics of SR-form, i.e., loss of virulence while preserving antigenic structure and immunogenicity seem to suggest the possibility of its application as a vaccine.
