Abstract


Aflatoxin is one of the mycotoxins, which can be extractable from some species of Aspergillus flavus, and it may cause acute hepatic periportal necrosis, bile duct proliferation and hepatoma in some animals.
The author performed histochemical studies to clarify the effect of Aflatoxin Bl on the livers of male albino rats weighing about 200 gm.
Aflatoxin Bl dissolved with dimethyl sulfoxide was injected singly into the peritoneum ling /kg and 6mg/kg to 70 rats. Each experimental group was made of 5 rats and sacrified at 1 day intervals from the first to sixth day after injection. Hematoxylin-eosin, periodic acid schiff, Best´s carmine, oil red O, alkaline phosphatase and succinic dehydrogenase stains were performed on frozen and formalin-fixed liver tissues. The results obtained were as follows:
1. The single injection of Aflatoxin B, caused degenerative changes, such as acidophilic
degeneration, nuclear pyknosis, vacuolar changes, and focal and periportal necrosis on
hematoxylin-eosin stain.
2. The coarse intracytoplasmic granules were diffusely observed in PAS and Best´s carmine stains with periportal sparing.
3. The variable sized fat positive granules were noted at the periportal region and extended to mid and central zone of the lobules gradually.
4. The coarse granules which were noted around the periportal area in alkaline phosphatase
stain were gradually increased and fine diffuse intracytoplasmic granules were also fcund.
5. The cells around the periportal areas were positively stained in succinic dehydrogenase
staining of the liver of control rats and more deeply stained at the same area of the
experimental groups.
6. Those changes were earlier and more prominent in 6 mg/kg injection group than 1 mg/kg group, and they seemed to be dose-dependent.
In summary, the author considered that aflatoxicosis of rat liver decreases glycogen metabolism, and increases lipid accumulation and the activities of alkaline phosphatase and succinic dehydrogenase.
